Abstract
A novel multifunctional delivery system based on calcium carbonate nanoparticles has been developed for combination radiotherapy of melanoma. Encapsulation protocols for a therapeutic compound have been successfully implemented, using potassium 1-amino-1,6-dioxo-6-(4-methoxyphenyl)-2-cyano-4-((3-(ethoxycarbonyl)-4,5,6,7-tetrahydrobenzo[b]thiophen-2-yl)amino)hexa-2,4-dien-3-olate, which has an antitumor effect, and the radioisotope radium-223 as an example. Controlled pH-dependent kinetics of drug release is demonstrated. The efficiency of radiolabeling with radium-223 was ~95.9 ± 3.1% with a high degree of activity retention in vitro (more than 90% after 144 h). The developed delivery system based on calcium carbonate nanoparticles demonstrated high biocompatibility with MDCK cells. The resulting radiopharmaceutical, in its “cold” modification, demonstrated dose-dependent cytotoxicity against B16-F10 melanoma cells, confirming the potential of this platform for subsequent use in combination radiotherapy for melanoma.
