Abstract
Objective: To assess the efficacy and safety of vitamin A and retinoid analogues for adult olfactory dysfunction.
Methods: A review conducted in accordance with the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines searched PubMed, Embase, Web of Science and the Cochrane Library from inception to September 2025 for clinical studies evaluating vitamin A or retinoids. Eligible designs included randomised trials, controlled trials, prospective cohorts and large case series.
Results: Eight studies including 589 participants were identified. Findings were inconsistent: observational studies and pilot trials suggested possible benefit, particularly in post-infectious disease and with olfactory training, whereas two randomised trials found no significant added benefit. Safety reporting was limited.
Conclusions: Current evidence does not support routine vitamin A treatment for olfactory dysfunction. Any benefit is uncertain and may be phenotype-, route- or dose-specific; adequately powered trials using standardised endpoints and adverse-event reporting are required.
