Abstract
This study aimed to investigate how Saposhnikoviae radix regulates skin barrier function. Bioinformatics analysis was conducted to identify the compounds in Saposhnikoviae radix and their target genes related to cell barrier function. Molecular docking analysis was performed to investigate the interaction between carvotanacetone (CARV) and key genes. The impact of CARV on skin barrier function was assessed in HaCaT cells treated with tumor necrosis factor-alpha (TNF-α) and interferon-gamma (IFN-γ). The effects of CARV and specificity protein 1 (SP1) on cell viability and permeability were analyzed. CARV effectively interacted with SP1. CARV attenuated the TNF-α/IFN-γ induced impairment of cell viability and Claudin-1 expression in an SP1-dependent manner, as SP1 knockdown abrogated its beneficial effects on Claudin-1 and cell permeability. CARV, a compound derived from Saposhnikoviae radix, protects HaCaT cells from barrier dysfunction by upregulating Claudin-1 expression via the SP1.
