Abstract
Excessive intake of kanamycin (KAN) poses significant risks to human health, so it is necessary to develop rapid, portable, low-cost, and highly sensitive biosensors to monitor KAN. Herein, a colorimetric/fluorescent bivalent aptasensor for rapid detection of KAN was developed based on bivalent aptamer-templated silver nanoparticles (BApt-templated AgNPs) and bivalent aptamer-templated silver nanoclusters (BApt-AgNCs), which was constructed by cDNA-BApt-cDNA, NaBH4 and AgNO3. BApt-templeted AgNPs and BApt-AgNCs can be synthesized by changing the reaction temperature and time of NaBH4 and AgNO3 with cDNA-BApt-cDNA. This aptasensor is composed of a colorimetric/fluorescence dual detection module, and it supports absorbance/RGB/fluorescence three-mode detection. An ultralow detection limit of 5.98 fM was achieved in the fluorescence detection module and a detection range of 0–500 μM was achieved in the colorimetric detection module. Meanwhile, the aptasensor demonstrates excellent repeatability in actual sample testing, thus having great potential for on-site quantitative analysis and portable detection of KAN.
