Abstract
Despite the notable success of traditional therapeutic approaches such as chemotherapy, significant limitations remain, including high dosage requirements, drug resistance, nonselectivity, adverse side effects, and poor bioavailability. To address these challenges, drug delivery systems (DDSs) have been developed to more effectively transport therapeutic agents to target sites while minimizing systemic toxicity. Among the strategies for designing smart DDSs, reversible covalent interactions, particularly Schiff base linkages, have emerged as crucial components due to their dynamic and stimuli-responsive properties. In this review, we emphasize the role of Schiff base chemistry in DDSs, highlighting its structural advantages, responsiveness to physiological triggers (e.g., pH), and its contribution to enhanced therapeutic efficacy. We also examine the integration of Schiff base compounds into various polymeric DDS architectures and discuss recent advancements in their applications for selective and controlled drug release. Finally, we consider the current challenges and future directions for achieving precise, targeted drug delivery through Schiff base-based systems.
