Abstract

Ionizing radiations, beyond their direct cytotoxic effect, may influence tumor immunogenicity. Studies conducted on preclinical models of alpha radioimmunotherapy (RIV-α) provide support for the hypothesis of potential immune system activation. However, documenting these immunological phenomena remains complex, as RIV-α differs from external radiotherapy in terms of dosimetry, making it difficult to establish a clear dose-response relationship. Additionally, the systemic nature of RIV-α complicates the clinical evaluation of its effects on non-target lesions. Among the important scientific challenges to be addressed in order to optimize the clinical efficacy of RIV-α are the identification of predictive biomarkers and the development of combined therapeutic strategies, particularly in association with immunotherapies.
