The proposed methods were validated as per the ICH guidelines parameters like Linearity, precision, accuracy, selectivity, limit of detection and limit of quantitation. 摘要

采用两种色谱法同时测定盐酸沙格列汀(SAG)和盐酸二甲双胍(MET)二元混合物的含量。第一种方法为反相高效液相色谱法。色谱分离采用Kinetex™色谱柱- c18 (4.6 × 150 mm, 2.6µm),流动相为乙腈:磷酸缓冲液pH = 4.5±0.1,正磷酸调节(13:87,v/v)。等温洗脱,流速1.5 mL/min,紫外检测波长220.0 nm。第二种方法是光谱密度法。色谱分离在60f254预涂硅胶铝板上进行,固定相为氯仿:甲醇:甲酸(80:20:0.3,体积)。用紫外检测器在210.0 nm处测定分离带的密度。按照ICH指南的线性度、精密度、准确度、选择性、检出限和定量限等参数进行验证。所得结果与文献报道方法所得结果进行统计比较,在准确度和精密度上均无显著差异。

Chromatographic separation was done on precoated silica gel aluminium plates 60 F254 as a stationary phase and developing system consisting of chloroform:methanol:formic acid (80:20:0.3, by volume).
