Pathophysiological mechanisms have been elucidated in the past decades. First of all, AD is progressive leading to cognitive deficits till dementia. Pathologically, AD features synaptic dysfunction with changes of neuronal circuitry, progressive accumulation of protein aggregates such as the beta amyloid and tau.
