Mucoadhesivebuccal films of candesartan were prepared by solvent casting technique using chitosan, HPMC, gelatin and EDTA as permeation enhancer. Prepared buccal films evaluated for various pharmaceutical parameters, stability studies, in-vitro and ex-vivo evaluation parameters performed. In-vitroangiotensin II receptor antagonist studies were also performed. Results showed improved bioavaibility of candesartan through buccal films. 摘要

坎地沙坦是有效的血管紧张素II受体拮抗剂类降压药物。但其水溶性差,首过代谢广泛。本文主要研究坎地沙坦口腔膜的研制。由设计专家对口腔膜进行了优化。选择了最佳的浓度范围,用于开发坎地沙檀颊膜的试验批次。以壳聚糖、HPMC、明胶和EDTA为渗透增强剂,采用溶剂铸造法制备坎地沙坦粘膜黏附口腔膜。制备的口腔膜对各种药物参数、稳定性研究、体外和离体评价参数进行了评估。也进行了体外血管紧张素II受体拮抗剂的研究。结果表明坎地沙坦通过口腔膜可提高生物利用度。关键词:坎地沙坦,血管紧张素II受体拮抗剂,颊膜,Box-Behnken设计Mucoadhesive力量。
