LPS was extracted by phenol hot water method and partial purified by gel filtration chromatography on sepharose CL-6B gel. The results showed that the percentage of carbohydrate amount in the partially purified LPS extract was 9.5% while the percentage of protein in the same extract was 0.06% with no nucleic acids. Intravenous administration of (0.1and1)μg/kg of partial purified LPS in the rabbit led to increase body temperature in dose dependent manner (38.8° and 40.2°)C respectively. Intraperitonial administration with the dose 10μg/mouse cause an increasein serum cytokine (IL-1α, IL-6 and TNFα) level after 4 hr of the injection. 摘要

从先前从新生儿血液样本中分离出的肺炎克雷伯菌侵袭性局部分离株中提取脂多糖。用苯酚热水法提取脂多糖,用凝胶过滤层析法对脂多糖CL-6B凝胶进行部分纯化。结果表明,部分纯化的LPS提取物中碳水化合物含量为9.5%,蛋白质含量为0.06%,且不含核酸。经静脉注射(0.1和1)μg/kg的部分纯化LPS可使兔体温呈剂量依赖性升高(分别为38.8°和40.2°)C。腹腔注射剂量为10μg/只,4小时后血清细胞因子(IL-1α、IL-6、TNFα)水平升高。结论:从肺炎克雷伯菌病原分离物中提取的高纯度LPS具有免疫调节作用,其作用机制是通过提高血清细胞因子(IL-6和IL-1α)水平,并在腹腔给药4hr后升高体温和tnf - α。
