According to the effect of different methods to HUVECs, HUVECs were divided into 3 groups: control group (containing 10% health serum medium), uremia group (containing 10% uremic patients serum medium), rhEPO group (medium containing 5, 10, 15 U/ml three different concentrations of rhEPO and 10% uremic patients serum medium medium) and 10% of patients with uremia. After Each medium of group culture effecting on endothelial cells 24 h, reactive oxygen species were detected by colorimetry and cell apoptosis was evaluated by TUNEL assay. \n \n \nResults \nrhEPO could reduce oxidative stress levels in cells, and had significant inhibition against apoptosis of endothelial cells induced by uremia serum (P<0.05), in addition, when the concentration of rhEPO increases, the oxidative stress levels and endothelial cell apoptosis index were reduced gradually (P<0.05). 摘要

目的rhEPO对尿毒症患者血清氧化应激水平及内皮细胞凋亡的影响。方法选取2015年1月~ 2016年1月新乡市第三附属医院尿毒症血清患者9例及健康志愿者9例。根据不同方法对HUVECs的影响,将HUVECs分为3组:对照组(含10%健康血清培养基)、尿毒症组(含10%尿毒症患者血清培养基)、rhEPO组(培养基中含有5、10、15 U/ml三种不同浓度的rhEPO和10%尿毒症患者血清培养基)和10%尿毒症患者血清培养基。各组培养基对内皮细胞作用24 h后,采用比色法检测活性氧含量,TUNEL法检测细胞凋亡情况。结果rhEPO能降低细胞氧化应激水平,对尿毒症血清诱导的内皮细胞凋亡有显著抑制作用(P<0.05),且随着rhEPO浓度的升高,氧化应激水平和内皮细胞凋亡指数逐渐降低(P<0.05)。方差相关分析结果显示,氧化应激水平与内皮细胞凋亡指数呈显著正相关关系(r=0.89, P<0.01)。结论rhEPO能明显抑制尿毒症血清诱导的内皮细胞凋亡,其机制可能与rhEPO能减轻氧化应激反应有关。关键词:重组人促红细胞生成素;尿毒症;氧化应激水平;内皮细胞;比色法
