Моноаминоксидаза как потенциальный биологический маркер эффективности терапии психических заболеваний
Conformational changes of the proteins participating in metabolic processes in human body may indicate the presence of serious pathological changes in it. The present work is devoted to evaluation of the state of patients with mental disorders by analyzing conformational changes of human serum albumin (HSA) by fluorescent methods. HSA is the important transport protein of blood plasma, binding various ligands, formed at metabolic processes. Changes in the structure of albumin molecule was detected by measuring of decay of fluorescent probe, K-35, bound to albumin globule and acting as a ligand-reporter. The measurements were carried out on serum albumin of the patients relating to different groups of mental pathology (29 patients with depression, 8 patients with schizophrenia). The data obtained while measuring serum albumin fluorescence for 7 healthy donors, were used as a control. It was shown, that the applied fluorescent method allowed us to distinguish statistically significantly average values of the parameters characterizing three time components of K-35 probe fluorescence decay for sick patients and healthy donors. The amplitude analysis of K-35 decay in fluorescence quenching (by nitrate anions) and ionic strength changes allowed us to differentiate the patients before and after treatment. Statistics of measurements collected so far is not sufficient to make early prognosis of the efficiency of therapy in the framework of the suggested fluorescent method. To prove the clinical significance of the obtained data the further investigation is carried out.
