Furthermore, the UPGMA dendrogram revealed that when sample from Guangnan in Yunnan province was used as one parent, reciprocal hybrids grouped with them in first, and then grouped with the other parent. The results indicated that the hybrids were closer to D. officinale from Guangnan population. This study identified the wild populations and hybrids of D. officinale by TRAP molecular markers, which is useful in selection of good varieties for artificial cultivation and early identification of hybrids. 摘要

本研究基于磷酸烯醇丙酮酸羧化酶(PEPC)、udp -葡萄糖焦磷酸化酶(UGP)和苯丙氨酸解氨酶(PAL)基因序列,利用TRAP分子标记对铁皮石斛野生居群和杂交进行了鉴定。从54个引物组合中选择7个多态性靶区扩增多态性(TRAP)引物用于野生群体的鉴定。杂交种具有雌性多态带、雄性多态带和杂合带,表明有7个TRAP标记能够从亲本中识别杂交种。UPGMA树状图显示,以云南广南为亲本时,互交杂交种先与之组合,再与另一亲本组合。结果表明,杂交种与广南种群的铁皮石斛较为接近。本研究利用TRAP分子标记对铁皮石斛野生居群和杂交种进行鉴定,为选育人工栽培良种和杂种早期鉴定提供依据。本研究为铁皮石斛种质稳定性和品质控制提供了一种方法。
