Gene sequencing, antigenic analysis and growth property were used to evaluate the rescued virus.</p><p><strong>Results: </strong>Rescued virus show the genes sequence correct, keep the same antigenicity and similar growth property compared with wild type virus.</p><p><strong>Conclusion: </strong>The pandemic virus reverse genetics platform of A/California/07/2009 (H1N1) were built. 摘要

目的:利用反向遗传学技术构建甲型H1N1流感大流行毒株A/California/07/2009 (H1N1)的基因表达平台。方法:扩增8个基因片段,用双向载体连接重组质粒,将重组质粒共转染293 T细胞,拯救病毒。利用基因测序、抗原分析和生长特性对获救病毒进行评价。结果:获救病毒基因序列正确,与野生型病毒具有相同的抗原性和相近的生长特性。结论:A/California/07/2009 (H1N1)大流行病毒反向遗传平台已建立。基于该平台,获救病毒与野生型病毒在抗原性和生长能力上具有相似性。
