摘要

目的:研究1例艾滋病痴呆复合体(AIDS dementia complex, ADC)患者HIV-1 nef基因的遗传多样性,探讨ADC的氨基酸变异及其发病机制。方法:从1例死亡的ADC患者的脾脏和不同脑组织(基底节区、额叶灰质、脑膜、颞叶)中提取基因组DNA,采用PCR扩增nef基因。PCR产物克隆到pMD19-T载体上,经氨苄青霉素转化和蓝/白斑点筛选。对5个阳性克隆进行测序,并用BLAST进行确认。利用BioEdit和MEGA4对HIV-1 nef序列进行处理,得到Neighbor-Joining tree、p-Distances和ds/dn值。结果:不同组织分离的HIV-1 nef基因与HXB2相比存在遗传变异。此外,外周和大脑之间的部分变化是不同的。结论:从ADC患者提取的HIV-1 nef基因存在变异,且外周和中枢神经组织的变异不同,这些变异可能改变了nef的功能,有待进一步研究。
