Updates on Tau-Related Drug Targets and Potential Disease-Modifying Therapies for Alzheimer's Disease.
Alzheimer's disease (AD) is a chronic, multifactorial neurodegenerative disease affecting a large proportion of the elderly population with detrimental psychological and socioeconomic impacts. This review starts with a brief overview of the molecular aspects of AD. It aims to provide recent updates on several potential protein targets studied as valid therapeutic approaches such as Bassoon protein, triggering receptor expressed on myeloid cells 2 (TREM2), phosphorylated tau (chimeric molecules DEPTAC and PhosTAC), O-GlcNAcase (OGA), and misfolded protein seeds. This review emphasizes the importance of pursuing a multifaceted approach to diversify and synergize the current AD pharmacotherapy.
期刊介绍:
The American Journal of Pathology, official journal of the American Society for Investigative Pathology, published by Elsevier, Inc., seeks high-quality original research reports, reviews, and commentaries related to the molecular and cellular basis of disease. The editors will consider basic, translational, and clinical investigations that directly address mechanisms of pathogenesis or provide a foundation for future mechanistic inquiries. Examples of such foundational investigations include data mining, identification of biomarkers, molecular pathology, and discovery research. Foundational studies that incorporate deep learning and artificial intelligence are also welcome. High priority is given to studies of human disease and relevant experimental models using molecular, cellular, and organismal approaches.
