Therefore, current cell culture procedures substitute xeno-free culture media with added supplement serum for the traditional DMEM media with bovine fetal serum (FBS). This switch increased the production cost immensely and made it difficult to produce MSCs on an industrial scale. In this study, we optimized the condition of MSCs’ cultures by adjusting the amount of the supplement serum usage to reduce production costs for industrial manufacturing. 摘要

间充质干细胞(MSCs)由于其多谱系潜能和免疫调节能力已成为治疗免疫相关疾病的有效工具。然而,在临床实践中,高剂量的干细胞输注往往是必不可少的。因此,有必要在保证细胞质量的同时,生产足够数量的间充质干细胞用于人类临床应用。为了能够在患者身上使用干细胞,需要比使用不含来自假设的过敏机制的物质的无xeno培养基更严格的捕获程序。因此,目前的细胞培养方法是用添加补充血清的无异种培养基代替传统的含有牛胎血清(FBS)的DMEM培养基。这种转换极大地增加了生产成本,使得在工业规模上生产msc变得困难。在本研究中,我们通过调整补充血清的用量来优化MSCs的培养条件,以降低工业制造的生产成本。这是第一个声称减少无xeno补充血清的量对从越南儿童脐带分离的hUC-MSCs的质量没有影响的研究。
