Albumin content in separated samples was above 80% under the optimal operating condition. The contaminative membrane was cleaned with different methods. The best method was distilled water for 0.5 h, 250 ppm NaClO (35∼40 °C) alkaline solution for 1 h, then 0.2% SDS alkaline solution (35∼40 °C) for 1 h, and finally distilled water washing until neutrality. 摘要

首次采用膜技术分离鹿血白蛋白。微滤作为预处理方法,大大提高了超滤的分离效率。考察了超滤压力、温度、PH、料液总蛋白等因素对膜通量的影响。最佳操作条件为操作压力0.2 MPa,操作温度35℃,操作时间60 min, PH 6.3,料液总蛋白浓度23.8 mg/mL。在最佳操作条件下,分离样品的白蛋白含量可达80%以上。用不同的方法对污染膜进行清洗。最佳方法为蒸馏水0.5 h, 250 ppm NaClO(35 ~ 40°C)碱性溶液1 h,然后0.2% SDS碱性溶液(35 ~ 40°C) 1 h,最后蒸馏水洗涤至中性。膜通量良好,水通量回收率为94.7%。
