It uses an aminoallyl-modified nucleotide together with other dNTPs in a cDNA synthesis reaction with SuperScriptTM III Reverse Transcriptase. After a purification step to remove unincorporated nucleotides, the amino-fluorescent modified cDNA is coupled with a monoreactive, Nhydroxysuccinimide (NHS)-ester fluorescent dye.
