三瘤梭子鱼I型微卫星标记及其多态性鉴定
本研究采用生物信息学方法,从NCBI数据库中的13 985条est中筛选了三瘤波图努斯Ⅰ型微卫星。共鉴定出287个Ⅰ型微卫星,其中二核苷酸重复型微卫星最多173个,占鉴定微卫星标记的60.3%,三核苷酸重复型微卫星79个,占鉴定微卫星标记的27.5%,而六、五、四核苷酸重复型微卫星较少。在二核苷酸重复序列中,AC/GT型最多,占53.8%,AG/CT型次之,占37.0%,AT和GC较少。选取14个微卫星进行多态性分析,其中8个微卫星在三瘤假单胞菌野生居群中显示多态性,平均PIC值为0.57,杂合度为0.63。其中,6颗微卫星的PIC大于0.5,表现出较高的多态性。本研究表明,从EST数据中筛选Ⅰ型微卫星标记物的方法是可行的。这些Ⅰ型微卫星可用于三管棘球蚴遗传多样性、遗传作图和数量性状位点(qtl)鉴定的进一步研究。
