Amifostine enhanced the sensitivity of HL-60 cells to arsenic trioxide by downregulating the expression of Survivin,thus promoting apoptosis effect. Abstract

氨磷汀对HL-60细胞Survivin表达的影响
目的探讨三氧化二砷和氨磷汀诱导人急性早幼粒细胞白血病HL-60细胞体外凋亡的机制。方法分别用不同浓度的三氧化二砷和氨磷汀对HL-60细胞进行处理。MTT法测定细胞的抑制率。半定量RT-PCR检测Survivin的表达。结果三氧化二砷对HL-60细胞的增殖作用随着剂量的增加而下降,且三氧化二砷联合氨磷汀对细胞增殖的影响更明显,且三氧化二砷联合氨磷汀对HL-60细胞Survivin表达的抑制作用比单用三氧化二砷的作用更显著。结论三氧化二砷通过下调Survivin的表达诱导HL-60细胞凋亡。氨磷汀通过下调Survivin的表达增强HL-60细胞对三氧化二砷的敏感性,从而促进细胞凋亡作用。关键词:砷;Amifostine;Cytoprotection;HL-60细胞;生存素基因
