Abstract
of the poster by MEWIS et al. (2019) Helosciadium nodiflorum (L.) W.D.J.Koch (Syn. Apium nodiflorum L.), a wild relative of celery and celeriac, has been selected as a model species to promote the establishment of genetic reserves in Germany. The genetic diversity and geographic patterns of genetic differentiation was investigated in the frame of the model and demonstration project GE-Sell, financially supported by the German Federal Ministry of Food and Agriculture through the Federal Office for Agriculture and Food (grant number 2814BM110 to 112). H. nodiflorum is distributed more in the western and south western part of Germany, namely in the states Rheinland-Pfalz, Nordrhein-Westfalen, Saarland and Hessen. In total, 24 sites were selected to study the genetic diversity of the species, of which 20 have been finally investigated. A set of 17 polymorphic SSR markers were identified and used for the genetic analysis of differences in the H. nodiflorum occurrences. DNA was amplified by using PCR and Microsatellite (ABI) analysis of fragments was conducted by means of capillary electrophoresis. The observed heterozygosity (Ho) ranges between 0.043 and 0.543 and the expected (He) heterozygosity between 0.308 and 0.848. The measure ∆SD(j) was calculated to evaluate the impact of each occurrence to genetic differentiation. HnLOE (∆SD(HnLOE) = 0.2901) represents the genetic composition of the pooled remaining 19 occurrences best and HnROD (∆SD(HnROD) = 0.6754) deviates most from its complement. Based on molecular and other biological parameters 15 occurrences were selected for the establishment of genetic reserves. However, re-visiting the population sites after 12 month and monitoring of the area revealed that two populations were Genetische Erhaltungsgebiete für Wildpflanzen für Ernährung und Landwirtschaft – Fachtagung 2019 Julius-Kühn-Archiv, 466, 2020 99 strongly reduced in number and distribution area due to abiotic and biotic factors. Therefore, only 13 occurrences for most appropriate crop wild relative population could be recommended with emphasis.
