Single-cell RNA sequencing (scRNA-seq) has proven to be a powerful strategy to study the heterogeneity in clinical samples, providing an unbiased approach to uncover the characteristics in different cell subsets. To ensure the reproducibility and robustness of biological discoveries, researchers need to be aware of hidden caveats in tissue dissociation, cell capturing and transcripts measurement which may affect cell composition assessment and cellular function annotation.
