OBJECTIVE: This study was performed in order to determine if mathematical modeling of the canine teeth in dogs could be utilized to provide an accurate and reliable estimation of crown surface area that could be used in both a research and a clinical setting. MATERIALS AND METHODS: Actual surface area (aSA) calculations for 32 stone dies of clinical crown preparations were acquired utilizing a tridimensional (3D) laser scanner and 3D computer-aided design and manufacturing (CAD/ CAM) software applications. These calculations were used as a control. Seventeen unique mathematical models from eight geometric shapes were used to calculate estimated surface area (eSA) of each stone die. Linear association and agreement between eSA and aSA calculations were assessed with multiple statistical methods. RESULTS: All methods of eSA showed a significant linear association with aSA. Five of the mathematical models [right elliptical frustum (H3), right elliptical cone (G3), right pyramidal cone (A3), right circular frustum (F2), and right circular cone (E1)] were superior to the other 12 models. CONCLUSION: The H3 mathematical model based on the right elliptical frustum provided the most accurate estimate of crown surface area of dog teeth. However, H3 requires the use of laser scans and a 3D CAD software program. As a result, this model would be recommended for research applications. The E1 mathematical model was similar in accuracy to H3 and, given it requires only two measurements and a comparatively simple equation for calculation, this method would be recommended for clinical chair-side use. Many traumatically induced feline fractures have been reported to involve the mandible or skull and, in cats with maxillofacial trauma, traumatic dentoalveolar injuries are particularly prevalent. Traumatic dentoalveolar injuries can also often be found in otherwise healthy cats. CLINICAL CHALLENGES: Some traumatic dentoalveolar injuries require emergency treatment; timely recognition and management is therefore important for achieving the optimal outcome. Multiple approaches exist for the management and repair of maxillofacial traumatic injuries. However, those for traumatic dentoalveolar injuries may be more limited. The aim of this report was to characterize osteonecrosis of the jaws (ONJ) in previously irradiated fields in dogs that underwent radiotherapy (RT) for oral tumors. Osteoradionecrosis of the jaw (ORNJ) was further defined as osteonecrosis in a previously irradiated field in the absence of a tumor. Thirteen dogs clinically diagnosed with 15 ONJ lesions were included in this retrospective case series. Medical records were reviewed for: breed, sex, weight, and age of the patient, tumor type, location in the oral cavity and size, location of the ONJ, time from RT to ONJ onset, known duration of the ONJ, and tumor presence. Where available, histological assessment of tissues obtained from the primary tumor, and tissues obtained from the ONJ lesion, was performed, and computed tomographic (CT) images and dental radiographs were reviewed. RT and other treatment details were also reviewed. Twelve dogs developed ONJ in the area of the previously irradiated tumor or the jaw closest to the irradiated mucosal tumor. Recurrence of neoplasia was evident at the time of ONJ diagnosis in five dogs. Time from RT start to ONJ onset varied from 2 to 44 months. In three cases, ORNJ developed after dental extractions in the irradiated field. Mesenchymal stem cells (MSCs) are a promising therapy for immune-mediated and inflammatory disorders, because of their potent immunomodulatory properties. In this study, we investigated the use of fresh, autologous, adipose-derived MSCs (ASCs) for feline chronic gingivostomatitis (FCGS), a chronic, debilitating, idiopathic, oral mucosal inflammatory disease. Nine cats with refractory FCGS were enrolled in this pilot study. Each cat received 2 intravenous injections of 20 million autologous ASCs, 1 month apart. Oral biopsies were taken before and at 6 months after the first ASC injection. Blood immune cell subsets, serum protein, and cytokine levels were measured at 0, 1, 3, and 6 months after treatment to assess immunomodulatory effects. Seven of the 9 cats completed the study. Five cats responded to treatment by either complete clinical remission (n 1⁄4 3) or substantial clinical improvement (n 1⁄4 2). Two cats were nonresponders. Cats that responded to treatment also exhibited systemic immunomodulation demonstrated by decreased numbers of circulating CD8þ T cells, a normalization of the CD4/CD8 ratio, decreased neutrophil counts, and interferon-g and interleukin (IL)-1b concentration, and a temporary increase in serum IL-6 and tumor necrosis factor-a concentration. No clinical recurrence has occurred following complete clinical remission (follow-up of 6-24 months). In this study, cats with <15% cytotoxic CD8 T cells with low expression of CD8 (CD8lo) cells were 100% responsive to ASC therapy, whereas cats with >15% CD8lo cells were nonresponders. The relative absence of CD8lo cells may be a biomarker to predict response to ASC therapy, and may shed light on pathogenesis of FCGS and mechanisms by which ASCs decrease oral inflammation and affect T-cell phenotype. SIGNIFICANCE: This study is the first to demonstrate the safety and efficacy of fresh, autologous, adipose-derived stem cell systemic therapy for a naturally occurring, chronic inflammatory disease in cats. The findings demonstrate that this therapy resulted in complete clinical and histological resolution or reduction in clinical disease severity and immune modulation in most cats.
