The optimum condition for extraction were 20 mL methanol as the solvent, the weight of sample 1,5 g, and extraction time 60 minutes. The total antioxidant content from Kaempferia galanga L. rhizome extraction by maceration and UAE were 0.6688 ± 0.02 and 0.8126 ± 0.02 mg/g DW. This study also validated the method of determining the total antioxidant content extracted with ultrasonic. The results of the method validation obtained linearity (r) = -0.9984, LoD value = 0.0283 mg/L, LoQ value = 0.0947 mg/L, precision (SDR = 1.97% and HORRAT = 0.96) and percent recovery 103.21%. 摘要

山柰是一种传统的药物和调味品。良姜山柰具有多种抗氧化活性。浸渍法提取抗氧化剂耗时长,提取工艺不完善,因此采用超声辅助提取法加快提取过程。本研究提出用UAE法提取高良山柰中抗氧化剂,并采用1,1-二苯基-2-吡啶肼(DPPH)法测定抗氧化剂的含量。最佳提取条件为甲醇20 mL,样品质量1.5 g,提取时间60 min。浸渍法和UAE法提取的山柰根茎总抗氧化剂含量分别为0.6688±0.02和0.8126±0.02 mg/g DW。验证了超声提取法测定总抗氧化剂含量的方法。方法验证结果为线性(r) = -0.9984, LoD值= 0.0283 mg/L, LoQ值= 0.0947 mg/L,精密度(SDR = 1.97%, HORRAT = 0.96),回收率103.21%。基于该方法的验证值,可以认为利用UAE提取的根茎中总抗氧化剂含量的测定是可行的。
