Eight teeth were used to monitor biofilm formation and maturation over the incubation period with field emission scanning electron microscopy (Fe SEM), while the other 60 teeth were divided into two experimental groups (n=20) in which 2% chlorhexidine digluconate functionalized hydroxyapatite nanoparticles (Group A) and 0.02% silver nanoparticles (group B) were used as intra canal medications, and two control groups (n=10). The positive control group (Group C) was used to check for bacterial viability throughout the experiment, while the negative control group (Group D) was used to check for sterility of the procedures. Finally, bacterial samples were collected and analyzed quantitively by culture counts and qualitatively by real time PCR (RTQ-PCR). \nResults: RTQ-PCR detected E. faecalis DNA in all groups except for the negative control group. The mean values of E. faecalis DNA detected in groups A, B and C were 99.6 ng/mL,  67.1 ng/mL , 2797.4 ng/mL respectively. Statistical analysis of these results showed that both treatment groups presented statistically significantly lower mean values of E. faecalis DNA quantities compared to the positive control group. 摘要

目的:观察和比较二光酸氯己定功能化羟基磷灰石纳米颗粒和纳米银纳米颗粒作为根管内药物对拔牙根管内成熟粪肠球菌生物膜模型的抗菌和抗生物膜活性。方法:机械制备68颗人上颌中切牙,消毒后感染粪肠杆菌,在厌氧条件下培养28天,形成成熟的粪肠杆菌生物膜。8颗牙采用场发射扫描电镜(Fe SEM)监测培养期内生物膜的形成和成熟情况,其余60颗牙分为2个实验组(n=20)和2个对照组(n=10),其中以2%二光酸氯己定功能羟基磷灰石纳米颗粒(A组)和0.02%纳米银纳米颗粒(B组)作为管内用药。阳性对照组(C组)在整个实验过程中进行细菌活力检查,阴性对照组(D组)在整个实验过程中进行无菌检查。最后收集细菌样本,通过培养计数和实时荧光定量PCR (real - time PCR, RTQ-PCR)进行定量分析。结果:RTQ-PCR除阴性对照组外,其余各组均检测到粪肠球菌DNA。A、B、C组粪肠杆菌DNA检测平均值分别为99.6 ng/mL、67.1 ng/mL、2797.4 ng/mL。对这些结果进行统计分析表明,与阳性对照组相比,两个治疗组的粪肠球菌DNA数量平均值均有统计学意义显著降低。但两组间差异无统计学意义(P< 0.05)。结论:二光酸氯己定功能化羟基磷灰石纳米粒子和纳米银纳米粒子对成熟粪肠球菌生物膜具有有效的抗菌活性。
