Light-scattering polarimetric spectrum is an effective tool to measure these features for quantitative pathology analysis. In this paper, a theoretical model of single backscattering spectra based on Mie theory is established, to describe the polarized light scattering spectroscopy for a two-layered scattering medium. To test the utility of this model, an epithelial tissue polarimetric spectral imaging system is proposed, then human frostbite sample is imaged using the system. Proposed model is used to acquire the morphological parameters closely relating to the abnormality of biological tissue, including the mean diameter, size distribution and relative refractive index.
