M. indica leaves were extracted using soxhletation with methanol. The cytotoxic test was perfomed using MTT assay with various MEML concentrations: 15.625, 31.25, 62.5, 125, 250 and 1000 μg/mL. Meanwhile, Bax and Bcl-2 expressions of MCF-7 breast cancer cells were detected with immunocytochemical staining after treatment of MEML with concentrations of ¼ IC50, ½ IC50 and IC50. The results showed that IC50 of cytotoxic activity was 308.12 µg/mL. MEML could influence Bax and Bcl-2 protein expressions. 摘要

研究了芒果提取物对MCF-7细胞的细胞毒活性。本研究旨在检测籼米叶甲醇提取物(MEML)对MCF-7乳腺癌细胞的细胞毒活性,并探讨MEML对Bcl-2和Bax表达的影响。采用甲醇蒸馏法提取籼稻叶片。采用MTT法对不同浓度的MEML(15.625、31.25、62.5、125、250、1000 μg/mL)进行细胞毒试验。同时免疫细胞化学染色检测MEML在¼IC50、½IC50和IC50浓度下作用MCF-7乳腺癌细胞后Bax和Bcl-2的表达。结果表明,其细胞毒活性IC50为308.12µg/mL。MEML可影响Bax和Bcl-2蛋白的表达。经MEML处理后,Bcl-2表达降低,Bax表达升高。关键词:芒果,细胞毒性,Bax, Bcl-2, MCF-7
