Steenis) merupakan tanaman yang Allah ciptakan dan dimanfaatkan sebagai obat. Al Quran surat an Nahl ayat 11 menjelaskan bahwa Allah telah menciptakan berbagai tumbuhan dan memberikan manfaat bagi orang-orang yang memikirkannya. Penelitian ini bertujuan untuk mengidentifikasi flavonoid serta mengetahui pengaruh pemberian ekstrak etanol 70% umbi binahong terhadap kadar glukosa darah dan aktifitas SOD pada tikus (Rattus norvegicus) yang diinduksi aloksan. Metode yang digunakan untuk memperoleh ekstrak adalah maserasi dengan pelarut etanol 70 %. Kemudian ekstrak pekat yang diperoleh digunakan untuk uji golongan senyawa menggunakan uji fitokimia dan pengujian antidiabetes terhadap tikus yang diinduksi aloksan dosis 32 mg/200g BB. Ekstrak lalu dihidrolisis dan dipartisi dengan kloroform:air (1:1) serta dipisahkan senyawa flavonoidnya dengan kromatografi lapis tipis (KLT) analitik menggunakan variasi eluen. Hasil penelitian menunjukkan bahwa ekstrak etanol umbi binahong positif mengandung golongan senyawa flavonoid, alkaloid, tanin, saponin dan terpenoid. Pemisahan golongan senyawa flavonoid telah memberikan pemisahan paling baik dengan campuran eluen butanol: asam asetat: air (4:1:5) yang menghasilkan 6 spot. Sedangkan pengujian antidiabetes pada tiga macam dosis 25, 50 dan 75 mg/kg BB ekstrak umbi binahong telah memberikan pengaruh penurunan kadar glukosa darah dan peningkatan aktifitas SOD (Superoksida dismutase) berturut-turut 5,511; 5,217 dan 4,942 u/ml \n \nENGLISH: \n \nBinahong (Anredera cordifolia (Ten.) The aims of this research was to identify the flavonoid content in binahong tuber and determine the effect of 70 % ethanol extract of binahong tuber on blood glucose level and SOD activity in rats (Rattus norvegicus) induced alloxan. The method used to obtain the extract was maceration using ethanol 70 %. The concentrated extract obtained was used to test the classes of its compound content using phytochemical test and antidiabetic effect against rats induced alloxan with dose of alloxan was 32 mg / 200 g BW. Then the extract was hydrolyzed and partitioned with chloroform: water (1: 1). The flavonoid compound contained in the extract was separated by analytic thin layer chromatography (TLC) technique using various eluents. The results showed that the 70 % ethanol extract of binahong tuber contains flavonoids, alkaloids, tannins, saponins and terpenoids. Butanol: acetic acid: water (4: 1: 5) was the best eluent for separation of flavonoid compound in extract as it produce 6 spots.
