After successful modeling, the rats in the long-snake moxibustion treatment group underwent long-snake moxibustion treatment every other day (along the governor vessel from Dazhui (GV14) to Shenshu(BL23), for a period of 14 days. The remaining two groups were secured in the same way as the long-snake moxibustion treatment group, although they did not receive any treatment. The body weight, rectal temperature, and spontaneous activity count of the rats, as well as serum levels of corticotropin releasing hormone (CRH) and </span>corticosterone<span> (CORT) were detected by ELISA before modeling, after modeling, and after treatment. The amount of 11β-HSD1 protein in rat liver was determined by immunohistochemistry<span> and Western blot analysis.</span></span></span></p></div><div><h3>Results</h3><p>Compared with the rats in the blank control group, those in the model group exhibited a significant decrease in the trend of body weight growth and in rectal temperature (<em>P</em> &lt;0.05), as well as a slight yet non-significant decrease in spontaneous activity count (<em>P</em> &gt;0.05); compared with the rats in the model group, the rats in the treatment group exhibited a significant increase in rectal temperature (<em>P</em> &lt;0.05) and in spontaneous activity count (<em>P</em> &lt;0.05). 摘要

目的观察长蛇灸对肾阳虚大鼠下丘脑-垂体-肾上腺皮质(HPA)轴及肝脏11β-羟基类固醇脱氢酶1型(11β-HSD1)表达的影响,为后续深入探讨长蛇灸对肾阳虚大鼠的作用机制提供基础。方法选取spf级雄性SD大鼠15只,随机分为空白对照组、模型组和长蛇灸治疗组,每组5只。采用氢化可的松粉30 mg/kg灌胃制备肾阳虚大鼠模型,灌胃量为10 mL/kg。造模成功后,长蛇灸治疗组每隔一天(沿督脉从大椎(GV14)至肾俞(BL23))进行长蛇灸治疗,连续14天。其余两组与长蛇灸治疗组相同,但未接受任何治疗。采用ELISA法检测造模前、造模后、给药后大鼠的体重、直肠温度、自发活动计数及血清促肾上腺皮质激素释放激素(CRH)、皮质酮(CORT)水平。免疫组化和Western blot检测大鼠肝脏中11β-HSD1蛋白的含量。结果与空白对照组比较,模型组大鼠体重增长趋势和直肠温度显著下降(P >0.05),自发活动计数略有下降(P >0.05);与模型组比较,给药组大鼠直肠温度显著升高(P <0.05),自发活动计数显著升高(P <0.05)。治疗14 d后,与空白大鼠比较,模型组大鼠血清CORT含量显著降低(P <0.05),肝脏11β-HSD1表达显著降低(P <0.05),血清CRH含量略有降低(P >0.05);与模型组大鼠比较,治疗组大鼠血清CRH含量显著升高(P <0.05),肝脏11β-HSD1表达显著升高(P <0.05),血清CORT含量略有升高(P <0.05)。结论长蛇灸能提高肾阳虚大鼠的直肠温度和自发性活动计数,改善HPA轴功能,增加肝脏11β-HSD1的表达。
