The cystic fluid of these children was collected as the case group and the corresponding infected viscera or pericystic tissue as the control group, with 40 cases in each group. In vitro cultured protoscolice of hydatid cyst, four groups including control group, LIF siRNA group, LIF factor group and SMAD4 siRNA group were divided by inhibiting TGF-β/SMADs signal pathway. Each assay was performed in triplicate. The expression of TGF-β, SMAD2/3/4 and LIF were detected.</p><p><strong>Results: </strong>The results of the clinical trial showed that the contents of SMAD2 and SMAD3 were increased in the case group compared with the control group; the differences were statistically significant (P < 0.05). The expression levels of TGF-β, Smad4, and LIF increased in the case group compared with the control group; however, the differences were not statistically significant. The results of further in vitro experiments, the expression levels of TGF-β, SMAD 2/3/4, and LIF after adding siRNA to interfere with Smad4 decreased in the case group compared with the control group; the differences were statistically significant (P < 0.05). Compared with the control group, the expression levels of TGF-β, SMAD2/3/4, and LIF increased after treatment with added LIF in the case group, and the expression levels of TGF-β, SMAD2/3/4, and LIF decreased after adding siRNA to interfere with LIF in the case group; the differences were all statistically significant (P < 0.05).</p><p><strong>Conclusion: </strong>SMAD2 and SMAD3 have a certain clinical relevance with hydatidosis in young children. The LIF expression level may be related to the cystic transformation of protoscoleces. 摘要

目的:探讨转化生长因子β (TGF-β)、果蝇母亲抗十足截瘫蛋白基因(SMAD) 2/3/4、白血病抑制因子(LIF)与幼儿肝粒棘球蚴囊肿形成的相关性。方法:选取2020年1月至2021年6月新疆维吾尔自治区人民医院符合儿童包虫病诊断标准的患者40例作为研究对象。取患儿囊液为病例组,取患儿相应感染的脏器或囊周组织为对照组,每组40例。通过抑制TGF-β/SMADs信号通路,将体外培养的原棘球蚴分为对照组、LIF siRNA组、LIF因子组和SMAD4 siRNA组。每项测定均为三份。检测TGF-β、SMAD2/3/4、LIF的表达。结果:临床试验结果显示,病例组患者血清SMAD2、SMAD3含量较对照组升高;差异均有统计学意义(P < 0.05)。与对照组相比,病例组TGF-β、Smad4、LIF表达水平升高;然而,差异没有统计学意义。进一步体外实验结果显示,病例组与对照组相比,加入siRNA干扰Smad4后TGF-β、SMAD 2/3/4、LIF的表达水平降低;差异均有统计学意义(P < 0.05)。与对照组比较,病例组添加LIF治疗后TGF-β、SMAD2/3/4、LIF表达水平升高,添加siRNA干扰LIF治疗后TGF-β、SMAD2/3/4、LIF表达水平降低;差异均有统计学意义(P < 0.05)。结论:SMAD2和SMAD3与幼儿包虫病有一定的临床相关性。LIF的表达水平可能与原头节的囊性转化有关。提示TGF-β/Smads/LIF信号通路可能参与原头节囊肿形成过程;为幼儿多房棘球绦虫卵感染后寄生的防治提供了研究依据。
