摘要

目的:探讨ITGB5表达下调对瘢痕疙瘩成纤维细胞增殖的影响,阐明β5-整合素(ITGB5)在瘢痕疙瘩中的可能作用。方法:构建靶向ITGB5的慢病毒sh- rna表达载体,感染瘢痕疙瘩成纤维细胞,采用Western Blot检测ITGB5的表达,MTT法检测其增殖能力。结果:KFb组、LV-NC组和LV-KFb组ITGB5 mRNA及蛋白表达量分别为1.00±0.00、1.08±0.05、0.34±0.01和0.91±0.03、0.93±0.02、0.28±0.07。与LV-NC组和KFb组相比,LV-KFb组ITGB5 mRNA和蛋白表达量显著降低(P < 0.01)。与LV-NC组和KFb组相比,LV-KFb组48 h细胞增殖率显著降低(P < 0.01)。结论:ITGB5具有促进瘢痕疙瘩成纤维细胞增殖的作用。
