The tested group was interfered by Tlr4-mus-1567 RNA which had the best result confirmed by QPCR, cells interfered by Negative Control RNA as NC group, and normal cell as control. 目的:探讨RNA干扰TLR4信号通路对Kupffer细胞吞噬的影响。方法:观察RAW2647小鼠单核巨噬细胞白血病细胞。实验组用QPCR证实效果最好的tlr4 -mu -1567 RNA干扰,阴性对照RNA干扰的细胞为NC组,正常细胞为对照。对各组进行吞噬试验。结果:实验组吞噬百分率低于对照组(17.67% +/- 3.51% vs 32.00% +/- 3.00%, P < 0.01),吞噬指数低于对照组(46.33% +/- 7.51% vs 82.00% +/- 6.08%, P < 0.01)。结论:RNA干扰可降低Kupffer细胞的吞噬活性。
