Two hundred one-day-old male broilers, divided into five groups, were fed with basal diet (control group), 0.3 mg/kg AFB1 (AFB1 group), 0.3 mg/kg AFB1 + 0.2 mg/kg Se (+Se group I), 0.3 mg/kg AFB1 + 0.4 mg/kg Se (+Se group II) and 0.3 mg/kg AFB1 + 0.6 mg/kg Se (+Se group III), respectively. According to biochemical assays, AFB1 significantly decreased the activities of glutathione peroxidase, total superoxide dismutase, glutathione reductase, catalase and the level of glutathione hormone, while it increased the level of malondialdehyde. Moreover, AFB1 increased the percentage of apoptosis cells by flow cytometry and the occurrence of apoptotic cells by TUNEL assay. Simultaneous supplementation with sodium selenite restored these parameters to be close to those in control group. 本试验旨在探讨亚硒酸钠对黄曲霉毒素b1诱导的肉仔鸡脾脏氧化应激和细胞凋亡的保护作用。1日龄雄性肉仔鸡200只,随机分为5组,分别饲喂基础饲粮(对照组)、0.3 mg/kg AFB1 (AFB1组)、0.3 mg/kg AFB1 + 0.2 mg/kg Se (+Se组I)、0.3 mg/kg AFB1 + 0.4 mg/kg Se (+Se组II)和0.3 mg/kg AFB1 + 0.6 mg/kg Se (+Se组III)。生化试验结果显示,AFB1显著降低了谷胱甘肽过氧化物酶、总超氧化物歧化酶、谷胱甘肽还原酶、过氧化氢酶活性和谷胱甘肽激素水平,同时显著提高了丙二醛水平。流式细胞术显示AFB1增加了凋亡细胞的百分比,TUNEL实验显示AFB1增加了凋亡细胞的发生率。同时补充亚硒酸钠使这些参数恢复到接近对照组的水平。由此可见,亚硒酸钠通过抑制氧化应激和细胞过度凋亡,对afb1诱导的肉仔鸡脾毒性具有保护作用。
Protective effects of sodium selenite against aflatoxin B1-induced oxidative stress and apoptosis in broiler spleen.
