摘要

利用reca介导的大肠杆菌同源重组技术,敲除加州自噬多核多角体病毒(AcMNPV)的p74基因,代之以斜纹夜蛾多核多角体病毒(SpltMNPV)的p74基因。修饰后的p74位点未留下影响p74基因表达和功能的选择标记。在重组AcMNPV bacmid-polhSL74中,AcMNPV p74基因启动子直接控制SpltMNPV p74基因的表达。RT-PCR结果显示p74基因被转录。生物实验结果表明,重组病毒AcMNPV bacmidolhsl74不能侵染银蛾幼虫,表明p74基因具有种特异性。
