A total of 112 fish were divided into 4 treatments and distributed as follows: fish anesthetized with 250 mg L<sup>−1</sup> at a temperature of 23 °C (<i>n</i> = 42); fish anesthetized with 250 mg L<sup>−1</sup> at a temperature of 27 °C (<i>n</i> = 42); and for each temperature, a control group fish exposed to diluent solution (95% ethanol) (<i>n</i> = 14, 7 per temperature); and a negative control group fish (water) (<i>n</i> = 14, 7 per temperature). They were exposed to 10 min for both temperatures. Blood collection was performed after the anesthetic induction at each temperature to determine the hematocrit, hemoglobin concentration (Hb), red blood cell count (RBC), mean corpuscular volume (MCV), mean corpuscular hemoglobin concentration (MCHC), mean corpuscular hemoglobin (MCH), relative values of leukocytes, and absolute values of leukocytes and thrombocytes, in addition to evaluating plasma chloride and total calcium ions. The analysis of chemical composition by gas chromatography coupled with mass spectrometry (GC–MS) demonstrated methyl-chavicol (70.04%) and linalool (24.59%) as the major components of basil essential oil. Fish anesthetized at a temperature of 23 °C showed lower neutrophil counts (<i>p</i> &lt; 0.05) compared to the other tested groups. For the temperature of 27 °C, values of MCV, MCHC, and RBC decreased (<i>p</i> &lt; 0.05) following exposure to basil essential oil. The calcium ion showed a statistical difference (<i>p</i> &lt; 0.05) between the treatments about the water control group. It is concluded that basil essential oil presented a positive anesthetic effect.
