Investigation of the incidence of pain in patients who underwent knee arthroplasty in the study. We aimed to examine the effect of the *17 allele of the CYP2D6 genome on the post-op tramadol activity. Methods: 110 patients who underwent total knee arthroplasty in our service in Pamukkale University Faculty of Medicine, Department of Orthopedics and Traumatology, and 100 healthy individuals without complaints as the control group were included in this study. Tramadol 100 mg (contramal) was administered intravenously to each patient. The port-op 0-15-30-45-60.min VAS scores of the patients were recorded. Results: The mean age of the patients was 62.36 years. While 86.4% of the patients in our study were female, this rate was 46% in the control group. We found 3.65% (n=14) *17 carriers with the presence of *17 allele in the patient (n=7) and control groups (n=7). The VAS score of the patients in the *1/*1 group at VAS post-op 0th minute was 91.07. The VAS score of the patients in the *1/*17 group was 95.0. There was no statistically significant difference between genomes (p>0.050). Similarly post-op 15., 30., 45. And 60. Min There was no statistically significant difference between the genomes per (p>0.050). We observed a statistically significant decrease in the postoperative VAS score between 0-60 minutes in both groups in time-dependent variation (p=0.000).
