摘要
以燕麦木聚糖和桦木为底物,研究了最佳条件下endo-β-1,4- d木聚糖酶的动力学参数(KM、VMax和kCat)。底物浓度(b/v)在0.2 ~ 1.2%范围内变化时,内切β-1,4- d -木聚糖酶的水解。在酶的最佳温度40°C下,孵育时间的变化可达20小时,间隔4小时。结果表明,内源性β-1,4- d木聚糖酶对燕麦木聚糖和桦木的KM分别为4.10 mg/ml和0.681 mg/ml。VMax值,kCat值为0.28 mg/ml。果酱和1.7 x 10-3 s-1。而白桦木底物的VMax和kCat为0.117 mg/U/jam和7 × 10-4 s-1。本研究发现,内切-β-1,4- d -木聚糖酶可以水解不同溶解度的底物。
