However, considering that a substantial proportion of those patients eventually relapses, as well as the relatively limited performance of those agents in second-line settings, a deeper understanding of resistance mechanisms is needed for enhanced guidance for patients' therapeutic selection in the second-line setting and beyond. In this review, we highlight trastuzumab's (<i>HER2</i>-targeting agent) performance in patients with gastric or GEJ cancer, with insight into mechanisms of resistance.
