We discuss how inflammation shapes clonal output from hematopoietic stem cells, how inflammation alters the immune microenvironment in the bone marrow, and novel therapies aimed at targeting inflammation in myeloid disease.</p><p><strong>Significance: </strong>Inflammation is emerging as an important factor in myeloid malignancies.
