In order to isolate the MAR fragment from the halotolerant green alga Dunaliella salina, we created a library of randomly obtained MAR from D. salina. Firstly the intact nuclei were released using 0.5% Triton X-100, then purification was carried out by discontinuous centrifugation using 30% and 70% Percoll gradients. Histones of nuclear matrices were removed using 25mmol/L lithium dioodosalicylate, the DNAs not closely associated with the matrices were removed using restriction enzymes. Suzhou) leaves were investigated by immunogold-labeling electron microscopy. The results showed that Rubisco and RCA were mainly located in chloroplasts of mesophyll, guard cell of stomatal apparatus and parenchyma of vascular bundle. A high density of gold particles were localized preferentially to the chloroplast stroma. In contrast, there were no specific binding of gold particles detected in the cytoplasm, vacuole, mitochondria and other organelles. As infected by turnip mosaic virus, the density of gold particles decreased lightly in the abnormal chloroplasts and dropped by much lower (58.44% and 64.67% as that in the health chloroplasts respectively) in swollen chloroplasts. The germination of maize seeds was inhibited seriously by NaCl treatment, while exogenous GABA reduced the inhibition of NaCl on the seeds germination. Effects on SOD, POD and CAT activity of GABA were detected. 1-2 mmol/L GABA induced the increase of the activity of SOD, POD and CAT about 20%. As a result, cells of tail fins were successfully passed from generation to generation, and constructed cell lines. During the culture procedure, the passage cells were observed and their biological characteristics were analyzed, which consist of morphologic analysis, drawing of growth curve, detection of producing rate for clones, insight of adaptation to temperatures, calculation of chromosomes and investigation for thaw of frozen cells. According to the observation results, we found that (1) there is an increase trend of fibroblast. epithelia of rostral side and tail fin from common carp are the best materials for culturing cells; (2) the optimum culturing temperature is 27 degrees C; (3) there isn't any change in cell chromosomes, which still are normal diploid. However, with, the generations increasing, the mode of chromosome has an inclination of decrease in normal ranges. We inferred the donor cells from 4-10 generations are the best choice for nuclear transplanted experiments in order to guarantee their genetic stabilities and survival rates after transplanting the nuclei. And, the cells will grow extremely well when cultured for 48-72h at 27 degrees C. We suggest the temperature should be descended little by little involving in preserving cells with DMSO. Minghui 63). The results showed that the inner and outer plasma membrane of amyloplasts, the web-like passages among starch grains and the amorphous substance surrounding the amyloplasts exhibited high ATPase activity. ATPase activity products were present on the membranes of protein body I and protein body II, and on the vacuoles and vesicle-like structures surrounding protein bodies. Active products of ATPase were also located on the plasma membrane and cell wall of starchy endosperm cells, on the cell wall, plasma membrane, nucleus and plasmodesma of aleurone and subaleurone layer cells. According to the distribution pattern of active products of ATPase, we infer that the web-like passage in amyloplast may be the transporting channel for nutrients flowing into the amyloplast. The results showed that ginsenosides distributed mainly in the assimilating tissue and phloem parenchyma cells, very little in collenchyma, epidermis and phelloderm, and no coloration in xylem and pith parenchyma. The accumulation of gingsenosides in the leaf occupied first place, the stem came second, and the root was the lowest. The content of total gypenosides showed a changing trend from low to high, then to low, during the whole growing period from vegetative growth to florescence and fructescence, and withering period. The content in leaf is higher than that in stem, and of male is higher than of female. Lymphocytes were collected from peripheral blood by lymphocyte isolation liquid (density 1.088), and transferred into enucleated oocytes by intracytoplasmic injection. Two hours later, oocytes injected with donor cell nuclei were activated for 6 hours by treatment in modified M16 medium containing 10 mmol/L SrCl2, and then cocultured with mouse oviduct epithelial cell in mM16 medium. On early day 4 blastocysts were cultured on mouse embryonic fibroblasts in DMEM. After 4 days, ICMs ( inner cell mass) were trypsinized, seeded and cultured. Results showed that donor nuclei of lymphocyte could be reprogrammed to support the development of reconstructed embryos in vitro. The proportion of reconstructed oocytes (n = 312) that developed to 2-cell stage was 41.03%, while 9.29% and 1.92% of reconstructed oocytes developed to morula and blastocyst phase, respectively. In addition, 2 ICMs were isolated from reconstructed blastocysts, and the proportion of reconstructed oocytes developing to ICM was 0.64% (2/312). {"title":"[Cysteamine attenuates suppression of peripheral IL-2 and lymphocytic proliferation induced by ruminal and duodenal operations in goats].","authors":"Zan Ming Shen, Hong Mei Xie","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Sixteen castrated male goats were randomly allocated into two groups (Control n=8; Experiment n=8) to investigate the effects of cysteamine on surgery-induced cellular immune suppression. The experiment commenced with 13 d of pre-operation (pre-Op), which contained 3 d of roughage-fed period (RP) and 10 d of roughage + concentrate-fed period (RCP), followed by first operation (Op-1) and then the second operation (Op-2). The goats were allowed to have 14 d of recovery between two operations. The fistulas were fixed to rumen walls in Op-1 and to duodenal walls in Op-2. Cysteamine (15 mg x kg(-1) x BW x d(-1)) was added to diet and fed to goats in Exp. group since RCP of pre-Op through whole experimental period. Blood was sampled through jugular vein from goats of both groups at pre-feeding of the last day of RP and RCP, 8 d after Op-1, 4 d and 13 d after Op-2, respectively to determine serum concentration of interlukin-2 (IL-2), cortisol, as well as PHA-stimulated lymphocytic proliferation rate (SI). In pre-operative periods there were no significant differences in concentration of IL-2 (0.82+/-0.1 vs 1.03+/-0.22 microg l(-1)) and cortisol (20.48+/-4.52 vs 20.80+/-9.93 microg l(-1)), as well as lymphocytic SI (90.72+/-7.25 vs 129.22+/-18.59) between Control and Exp. groups. The concentrations of IL-2 (0.82+/-0.1 vs 0.73+/-0.08 vs 0.55+/-0.12 microg l(-1)) and lymphocytic SI (90.72+/-7.25 vs 49.31+/-6.4 vs 7.09+/-1.66) in Control were depressed acutely (P<0.05), but cortisol concentration elevated (20.48+/-4.52 vs 26.67+/-10.51 vs 32.33+/-3.29 microg l(-1)) in post-operative periods (8 d after Op-1 and 4 d after Op-2), compared with those in RCP. While they remained unaltered in Exp. group except a slight decrease of lymphocytic SI at 4 d after Op-2. The different responses of Control and Exp. groups to surgery stress led to a 80-90% higher of IL-2 and about 3 times greater of SI in Exp. than those in Control, which was contributed by cysteamine administration. Cotyledons at 4-day seedlings were explanted and cultured on MS medium supplemented with different combinations of hormones. In \"Yixuan\" cultivar, the plantlet regenerative frequency and shoots produced by per explant was as low as 10% and 0.4 respectively at all combinations tested when distal parts of cotyledons were inoculated. Whereas, maximum regenerative frequency of 100 percentage and 10.3 shoots regenerated from per explant were observed on MS medium supplemented with 5 mg/L BA and 0.1 mg/L IAA using basal parts of cotyledons as explants. Likewise, in \"Jingxinmuben\" cultivar, the highest regenerative frequency of 100 percentage and 6.9 shoots regenerated from per explant were obtained when proximal parts of cotyledons were cultured on MS medium containing 2 mg/L BA and 0.5 mg/L IAA. In the present study, adventitious buds were initiated after only 4 d in culture and intact plantlets were obtained in 40 d culture. {"title":"[Distribution and toxicity of cadmium in Hydrilla verticillata (L.f.) In this study, aquatic vascular plant Hydrilla verticillata (L.f.) Royle was cultivated in water containing elevated concentrations of cadmium (up to 10 mg/L) for 7 d, the accumulation, subcellular distribution, ultrastructural localization, chemical form, toxic effects on mineral nutrient absorption of cadmium, photosynthesis rate and respiration rate were studied. It was found that H. verticillata fronds was able to accumulate cadmium, the bioconcentration factor was 193-307; subcellular fraction analysis revealed that cadmium major accumulated in cell wall (61.66%-52.00%) with decrease trends, the content of Cd enhanced in soluble fraction and remained stable in organelles, with the augment of pollutant concentration. The Cd levels occurred in different parts of leaf cell with the following sequence: cell wall > soluble fraction > organelles. Ultrastructural localization of cadmium with sulfide-silver method showed that Cd appeared in cell wall, chloroplast, nucleus and vacuole. Sequential extraction indicated that the ratio of different cadmium chemical form was different markedly, of which NaCl extractable Cd was predominated as compared with the other 5 forms; which could be seen in the following order: F(NaCl) > F(HAc) >F(Water) > F(Ethanol) > F(HCl) > F(Residue). The mineral nutrient absorption was also affected by cadmium stress, it increased the absorption of Ca, Mn, Cu, and Fe; but reduced that of P and K. Cd had a strong inhibitive effect on photosynthesis rate and respiration rate. The results suggested that the toxic symptoms of plant showed an evident correlation between dose and effect; the ultrastructural damage was closely related to the distribution of Cd.
