Seven days after the subcutaneous injection of ISO (15 mg/kg) the hearts were perfusion fixed and 12 sections from each heart were stained (Masson's trichrome). ISO induced myocardial fibrosis was quantified at the light microscopic level according to established morphometric principles. Pulse rate and ST elevation were recorded by EEC (3 standard leads) before and after the ISO injection. Non-diabetic control animals showed marked fibrosis after ISO, but surprisingly the diabetic animals showed no fibrosis after ISO treatment. These findings were in accordance with an ISO induced ST elevation seen only among control animals although both groups showed the same degree of tachycardia. Insulin treatment prevented the protection against ISO and when streptozotocin was injected 24 h after the ISO a normal quantitative and qualitative appearance of the scar tissue was seen. It thus seems that streptozotocin diabetic rats are protected against the toxic effect of ISO, leaving the haemodynamic response unaffected. Which factor in the diabetic metabolism is responsible for the present phenomenon is not known, but a defect in the signal transmission from the beta-receptor to the adenylcyclase is suggested as a possible explanation.</p>","PeriodicalId":76799,"journal":{"name":"Virchows Archiv. The aneurysmal bone cyst was composed of four different types of stromal cells - fibroblasts, myofibroblasts, osteoblasts, and histiocytes - and osteo-clastlike multinucleated giant cells. The surface of blood spaces was devoid of specialized endothelium, which may explain the presence of large quantities of extravasated erythrocytes. Some histiocytes contained siderosomes. The malignant lesion consisted of two main types of stromal cells, of which one had electron lucent and the other electron dense cytoplasm. The stromal cells produced osteoid and the tumour was regarded as an osteosarcoma. The multinucleated giant cells resembled those observed in aneurysmal bone cysts, but the nuclei seemed to be more often spherical. It is concluded that irradiation of the aneurysmal bone cyst may cause sarcomatous transformation in a cell capable of producing osteoid.</p>","PeriodicalId":76799,"journal":{"name":"Virchows Archiv. The rats characteristically developed fibrinoid necrosis of the wall and marked cellular thickening of the intima and media of the arterioles and small arteries of the kidney, testicle, mesentery, adrenal gland, brain, etc. Parenchymal damage of the organs, secondary to the vascular alterations took place. There were no accumulations of lipids in the vascular lesions. Though stroke has been stressed as a characteristic clinical feature of the SHRSP rats, the cerebral lesions are different from those seen in ordinary cerebrovascular disease. Furthermore, many organs are involved. The overall vascular changes in the brain and other organs are consistent with those seen in malignant hypertension; the SHRSP rat is an excellent model of this disease.</p>","PeriodicalId":76799,"journal":{"name":"Virchows Archiv. The neoplastic cells contained abundant mitochondria, moderate to large amounts of rough endoplasmic reticulum (RER) arranged in parallel arrays and often containing amorphous material, annulate lamellae, possible secretory vesicles, and glycogen. The presence of stacked RER and annulate lamellae together is unusual in papillary serous cystadenocarcinoma of the ovary, and has not been described in Fallopian tube adenocarcinoma. Golgi complexes were are. Small acini with projecting microvilli as well as junctional complexes were present, but cilia were not found. The electron microscopic findings suggest secretory activity, and are remarkably similar to those found in papillary serous cystadenocarcinomas of the ovary. The findings also support the hypothesis that ovarian serous tumors and adenocarcinomas of the Fallopian tube are derived from coelomic epithelium.</p>","PeriodicalId":76799,"journal":{"name":"Virchows Archiv. Forty-three were classified by the WHO scheme. Keratin immunoreactive cells were identified in all 8 epidermoid carcinomas; 6 out of 12 large cell carcinomas; 2 out of 6 adenocarcinomas; 2 out of 15 small cell carcinomas and in the only muco-epidermoid carcinoma. Using an immunohistochemical stain of tissue sections it has been shown that many of the antigenic determinants detected by the different antisera are identical. A radioimmunoassay for epithelial membrane antigen (EMA) showed that casein preparations are associated with small quantities of EMA. One of these tumors )(case 1) could be classified as a highly differentiated mucoid TSH cell adenoma presenting histochemical reactions typical of, as well as electron microscopical features identical to, normal TSH cells. Immunoenzymatic studies failed to dfemonstrate TSH in the tumor cells. Two further adenomas (case 2 and 3) were similarly structured in many areas, but showed regions of poorer differentiation in which distinct pleomorphism, irregular secretory granules, increased numbers of ribosomesand a well developed rough endoplasmic reticulum were present. In 10% of the tumor cells GH could be demonstrated immunoenzymatically, but there was no TSH. The fourth adenoma was an undifferentiated acidophilic adenoma showing pleomorphic cells having slight acidophil and partly mucoid granulations. The ultrastructure showed convoluted nuclei, increased numbers of free ribosomes as well as abundant rough endoplasmic reticulum and secretory granules which were differentin size and number but distinctly of the TSH cell type. Immunoenzymatically, TSH was found in some cells, with GH in more cells. Endocrinologically, elevated levels of GH were measured in cases 2, 3 and 4 with LH being increased in case 1. The amount of AAT remains unchanged during the morphogenetic stages of the pathological process (early, granulomatous, fibrosing phases), and does not correlate with the number or the presence of Michaelis-Gutmann (M.G.) bodies. Macrophages from other pathological processes, closely resembling malakoplakia cells but without M.G. bodies, did not contain AAT, except for a few macrophages in tuberculosis and xanthogranulomatous pyelonephritis. Whatever the source and the pathogenic role of AAT in malakoplakia, its presence in all macrophages seems to be specific for this disease. Immunohistochemical staining for AAT is therefore proposed as a useful test for an early and accurate differential diagnosis of malakoplakia.</p>","PeriodicalId":76799,"journal":{"name":"Virchows Archiv. The disease usually affects females, the clinical picture being dominated by severe disturbances of conduction. The original designations of the disorder stem from the morphological resemblance of the foamy cells to lipid-laden histiocytes. However, subsequent investigations have shown these cells to contain myofibrils interposed with Z lines. It has, therefore, been suspected that the leading cell population might be related to the myocardium. Using a histochemical method for the demonstration of cholinesterase activity in the foamy cells, we present evidence that \"histiocytoid\" cardiomyopathy may in fact correspond to a maldevelopment of the Purkinje cell system of the heart.</p>","PeriodicalId":76799,"journal":{"name":"Virchows Archiv.
