{"title":"[Pituitary-adrenal cortical function in newborn infants--especially in the ratio of unbound unconjugated corticosteroids (author's transl)].","authors":"T Oki","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>It has already generally been accepted that there are a variety of fetal adaptations to a stress during delivery and the fetal endocrinological functioning is being controlled mainly by the pituitary adrenal axis. An endocrinological difference between fetuses given a stress and the controls was observed in this study. The levels of ACTH and cds in the umbilical cord blood are supposed to be the indicators of stresses. The measurements of not only those levels but the percentile value of non-protein bound cds (NPC), known for an active form, were conducted under various conditions. Consequently, the three following results were obtained. 1. In the group of infants delivered by the vacuum extraction, the levels of ACTH and percent free cds tended to be more increased, and those of total cds and NPC were significantly higher than both groups of infants delivered spontaneously and by the oxytocin induction. 2. There were no significant differences in the levels of ACTH, total cds, percent free cds and NPC in plasma between the groups of mothers when delivered spontaneously and by the vacuum extraction. 3. {"title":"[Study on hyperprolactinemic anovulatory syndrome and treatment with bromocriptine (author's transl)].","authors":"T Fukunaga","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Clinical characteristic of the hyperprolactinemia were studied in 101 cases and effects of Bromocriptine were investigated in 57 cases with hyperprolactinemic anovulation. 1. Lactation was observed in 93.1% of the cases with hyperprolactinemia. There was no relationship between the degree of lactation and Prolactin (PRL) levels. 2. Hyperprolactinemia had no influence on basal gonadotropin levels. 3. In hyperprolactinemic anovulatory syndrome, a negative correlation between estradiol and PRL levels was found. PRL levels in the second grade amenorrhea were higher than those in other grade. 4. In LH-RH test, there was no relationship between PRL and LH-RH test. 5. With Bromocriptine therapy, ovulation was induced in 45 cases and pregnancy was successful in 31 cases. 15 cases of pregnancies were delivered of normal babies and 3 cases fell into abortion. 6. The initial dose of Bromocriptine was 2.5 mg/day. The dosage was increased by determining the PRL as index. 7. Cases with severe anovulation and high PRL levels, required prolonged administration. 8. 15 cases who had no ovulation by Bromocriptine administration for one month, ovulated successfully by clomiphene combined with Bromocriptine. 9. {"title":"[The occurrence rate of neonatal anomaly related to aging of grafted ovaries in mice (author's transl)].","authors":"Y Okada, K Ichinoe","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>An increase in the birth rate of malformed neonates with the aging of mothers has recently become clear. No studies, however, have been made as to whether such frequent occurrence of anomalies does depend on \"the aging of ovaries\", as has generally been assumed. The authors, in an attempt to elucidate the above mentioned basic subject, studied the relationship between the aging of ovaries and the occurrence rate of malformed newborn experimentally with C57BL/6J strain mice by grafting young castrated female mice orthotopically with different aged ovaries, from young to old, and mating them with young males. As controls, non-treated different aged females were mated with young fixed age males. The frequency in the birth of malformed mice in both the experimental group and the control group were examined and compared. As the age exceeded 400 days, the occurrence rate of anomaly rose sharply in the non-treated natural controls. When the grafted ovarian age exceeded 400 days, exactly the same result was obtained in the experimental group which showed a high incidence of neonatal anomaly. Recently, these experimental nodules were considered to be simulating the disseminated pattern of Leiomyomatosis Peritonealis Disseminata (LPD). Although these experimental nodules were considered to be composed of fibroblasts by light-microscopic studies, LPD was verified to be composed of smooth muscle by electronmicroscopic studies. Therefore, it is indispensable to study these experimental nodules by electronmicroscopy, when we discuss about the similarities between experimental nodules and LPD. Guinea pigs were treated with various doses of estradiol benzoate (E) [40, 80, 100, 200 microgram/day, thrice a week, intramuscular injection]. After treatment for three months, every dose of E produced the lesions simulating the disseminated pattern of LPD: multiple nodules in various sizes, scattered on the surface of the uterine horn, peritoneum, spleen and pancreas, and in the omentum and mesocolon. Ultrastructurally, the nodules were composed of the cells resembling fibroblast. The following conclusions could be drawn from them. (1) A low-dose irradiation on the primary cancer-nest intensified the interstitial reaction. (2) When PS-K was administered after low-dose irradiation, the interstitial reaction was intensified more remarkably than with irradiation alone. Moreover, it was possible for PS-K administration to restore the responsiveness of lymph nodes which had been inhibited by the irradiation. (3) Some of the observed morphological changes suggested the local occurrence of an immunological reaction. The following findings could be pointed out: (a) In the primary lesion of the uterine cervix, exudative cells consisting mainly of lymphocytes, plasma cells, and macrophages formed a barrier-like layer surrounding the cancer-nest. Granuloma accompanied by foreign-body type giant cells were formed in the interstitial tissue. Moreover, lymphoid tissues with a germinal center were formed. (b) In lymph nodes of the region involved, responsiveness was enhanced in the paracortical area, or a germinal center formed with its responsiveness enhanced. (4) There was a correlation between the degree of interstitial reaction of the primary lesion and the survival results of a follow-up survey. {"title":"Effects of conjugated estrogens on ripening of uterine cervix in nonpregnant castrated and pregnant rats.","authors":"Y Saito, S Takahashi, M Maki","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The stimulating effects of conjugated estrogens (CE, Premarin) on ripening of the uterine cervix were investigated by measuring the wet weight of the cervices, as well as by both light and electron microscopic examinations on non-pregnant castrated (hereinafter called castrated) and pregnant rats. Increase in the wet weight of the cervix was observed, in comparison with lactose control group, 4 and 2 hours after administration of CE in castrated and late pregnant rats, respectively. The maximum increase in the wet weight of the cervix was evident during the period of 4 to 10 hours after administration of CE. Scanning electron microscopic investigation was carried out on the cervices using the previously defined \"ripening score\". The scores of castrated rats cervices taken 4 hours after administration of different amounts of CE were 24.8 +/- 1.0 (n=5, 100 microgram CE/rat) and 24.8 +/- 2.0 (n=5, 100 microgram CE/rat), showing marked increases in score, as compared with those of control. The light and transmission electron microscopic figures of the cervices of castrated rats treated with CE resembled strikingly to those of pregnant rats, and the resemblance was also striking as compared with those of castrated rats treated with free estrogens. The onset of the action of CE on the cervical capillaries was apparently faster than that on cell components of the connective tissue. There was no essential difference in ultrastructure between the cervices of lactose treated pregnant rats and CE treated pregnant ones. CE exhibited more prompt effects in increasing the wet weight of the cervix than did free estrogens in castrated rats (as observed in earlier experiments).
