{"title":"[Regulation of cell growth exemplified by T lymphocytes].","authors":"C Hückel","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The aim of this paper is to describe the complexicity of growth regulation, particular of T-lymphocytes. Different surface molecules are known, which trigger the process of cell activation after binding of the respective ligands, including the T-cell-receptor/CD3 complex, the CD2 and the CD4 molecules. This surface molecules induce among other cellular events the expression of additional receptors, such as IL-2-receptors and Transferrin-receptors. The structure of these surface molecules, their possible functions and involved mechanisms of signal transmission from the membrane to the level of gene expression are presented. The different mechanisms of signal transmission and structures involved, such as the G-protein, are discussed. The different systems of signal transmission, in which such enzymes like Ca2+-dependent neutral Protease, Phospholipase C, Adenylatcyclase, Phospholipase A2 and Proteinkinases are involved, can interact. A relative deficiency in blood group O was found, with a shift toward an over-represented B-phenotype as compared to the general population. This shift appeared to be largely due to the contribution from the female patients with pollinosis. {"title":"[Novel immunotherapeutic models for neoplastic diseases of the urogenital tract with special attention to bladder carcinoma].","authors":"P Leskovar","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>In this review article some novel immunotherapeutic models as well as some new concepts concerning the unspecific and specific T-cell stimulation are discussed briefly. Some of these immunotherapeutic models are restricted to bladder cancer; the others have a generalized meaning. One model, restricted to bladder cancer, is based on the enzymatic++ pretreatment of effector-cells (macrophages, NK-cells), followed by their instillation in the patient's bladder. The other bladder tumor restricted model includes the direct in situ activation of effector cells by proteases and lipases. A third model for the treatment of bladder tumor implies a presensitization of patient's TD/TDTH-cells, followed by the treatment of patient's bladder tumor cells by some, common haptenic group. In addition, some novel ways of immune stimulation, partly by circumventing the (tumor-specific) immune tolerance, based on the cross-linking of crucial membrane structures on T4- and T8-cells or on a managed, controlled APC: T-cell interaction, are discussed. The Ig-producing initial cell lines were selected, and the supernatants were further analyzed for specific antigen binding (ELISA). IgM-antibodies were found which reacted with self- and non-self antigens of different molecular origin (nucleotides, proteins, carbohydrate structures). These antibodies were called multireactive (multispecific). The multispecific IgM-producing human hybridomas occurred with higher frequencies in the spleen (6.9% of IgM-producers) cell fusions than in experiments where peripheral blood-derived lymphocytes were fused (2.7%). There were no hybridomas producing multireactive antibodies detected in fusion material from lymph nodes. The greatest number of multireactive IgM was seen when PBL from SLE or anti-HIV-positive patients were hybridized to CB-F7 cells. Representative cell lines were cloned and recloned. The cells can be observed throughout the process. They are not exposed to mechanical stress after the fusion pulse. Electrofusion between the heteromyeloma line CB-F7 and human mononuclear cells from peripheral blood of immunized donors is shown to provide stable hybridomas producing IgG against tetanustoxin. It was shown that human hybridoma cells could be frozen most successfully at a cooling rate of 5 K/min whereas mouse hybridomas at 1 K/min. The percentage of FCS in cryopreservation media (30 or 90%) had no influence on the recovery of both types of hybridomas. For testing the efficiency of freezing, the fusion rate (number of growing and Ig producing hybridomas), the ratio of IgM:IgG-producing initial hybridoma lines and yields of specific wells were analysed. Comparable results were registered when optimally cryopreserved and non-frozen material was studied. The monoclonal antibody BL-Ac/p26 binds to an antigen which is expressed on activated lymphocytes. This antigen is not detectable on resting lymphocytes or other blood cells. The surface radioiodinated antigen of activated T cells isolated by the monoclonal antibody BL-Ac/p26 is separated by SDS-PAGE in a major 26k Da component and a minor band (32 kDa) under reducing conditions. These polypeptide chains form disulphide linked dimers on the cell surface (Mr 55-77 kDa). This 26 kDa antigen is a very early activation marker of T lymphocytes. {"title":"[Clinical relevance of determining lymphocyte subpopulations in peripheral blood of patients with rheumatoid arthritis].","authors":"M Hempel,&nbsp;F Wietschel,&nbsp;K Malberg,&nbsp;P Kästner","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Blood lymphocytes were determined from 21 patients with seropositive rheumatoid arthritis (RA) in a cross-sectional study with 5 monoclonal antibodies (MAb) of the BL-series. 7 patients were examined with the same MAb over a time period of 14 weeks before and after a prednisolone-pulse therapy. The cross-sectional study showed a significant higher percentage of Ia4+ T-helper/inducer-lymphocytes in RA-patients compared to control persons. The value of Ia4+ T-lymphocytes didn't correlate to the clinical activity of the RA. No significant differences were observed in the helper/suppressor T-lymphocyte ratio. The lymphocyte subpopulations were not significantly different immediately before and after a prednisolone-pulse therapy. The percentage of B-lymphocytes was significantly higher in comparison to the controls 24 h after prednisolone-pulse therapy.
