Deng, Xiao-Ying Hu","doi":"10.1093/mictod/qaad019","DOIUrl":"https://doi.org/10.1093/mictod/qaad019","url":null,"abstract":"\u0000 We describe a method for cleaning leaf epicuticular wax and contaminants from herbarium specimens in preparation for scanning electron microscopy. Specimens are rinsed in chloroform, heated at 50°C for 8 hrs, ultrasonicated in 70% ethanol, and subjected to critical-point drying. This method helps preserve and reveal the surface details of specimens that are heavily covered with epicuticular wax and contaminants, making many morphologically and functionally important details readily visible. Using cryoEM to determine structures of biomolecules at high resolution is a rapidly evolving technique with high barriers to entry for non-microscopists. To help academic researchers remain competitive in their fields, the NIH established a network of centers to broaden access and training in cryoEM. One initiative from this network is the monthly CryoEM Current Practices webinar that provides practical methodological tips and tricks and is openly accessible to anyone interested. {"title":"Scanning Transmission Electron Microscopy Done Right","authors":"","doi":"10.1093/mictod/qaad009","DOIUrl":"https://doi.org/10.1093/mictod/qaad009","url":null,"abstract":"\u0000 Performance, usability, and even availability of 4D-scanning transmission electron microscopy (STEM) measurements have historically been compromised by the lack of integration of 4D-STEM components on (S)TEM columns that were not originally designed for this purpose. This article introduces TESCAN TENSOR, the world’s first dedicated 4D-STEM instrument for multimodal characterization of nanoscale morphological, chemical, and structural properties of functional materials, thin films, and natural and synthetic crystals with stand-out 4D-STEM performance and unprecedented usability. 4D-STEM is the microscopy method of choice for nanoscale, multimodal characterization of material properties such as morphology, chemistry, and structure. At each pixel in the STEM dataset, the TENSOR rapidly acquires a diffraction pattern and an energy dispersive spectroscopy (EDS) spectrum that are perfectly synchronized. Together, diffraction and spectroscopy data encapsulate the full picture of electron beam–specimen interaction, from which a wide range of material properties can be derived. To make 4D STEM experiments as viable as conventional STEM image acquisition, the recording of diffraction patterns with a pixelated detector at fast frame rates, sufficient sensitivity to capture single electron hits, and high dynamic range is necessary. This paper addresses the recent development in hybrid-pixel detector technology that now allows 4D STEM experiments with a similar setup to conventional STEM imaging with pixel collection time under 10 µs.
