Accordingly, the phosphate dependent metabolisms of the soil-living streptomycetes should be stamped by lack of phosphate, too. The biosynthesis of the streptothricin antibiotic nourseothricin by Streptomyces noursei 3890b is initiated by limitation of soluble phosphate in the fermentation medium. At the other side is shown that a certain rate of feeding of phosphate during the fermentation increases the nourseothricin biosynthesis. An ambivalente role of phosphate on the secondary metabolite biosynthesis is stated. The limitation of phosphate leads to a special physiological state of the producer, characterized by secondary product formation and dephosphorylating activities in cells. This state is temporally stabilized by the presence of a sufficient phosphate supply, realized by enzymatic hydrolysis of complex phosphate-containing substrates or by a direct feeding of inorganic phosphate to the fermentations. The occurrence of different physiological states in respect to the phosphate-dependent metabolism is described by S-shaped functions of the relationship between specific growth rate and the phosphate concentration in the medium. The special behaviour of Streptomyces noursei cells at phosphate limitation is discussed to be the result of the dephosphorylating activities in cells, hydrolyzing phosphoester-bonds of regulatory metabolites as well as energy-rich compounds.</p>","PeriodicalId":23821,"journal":{"name":"Zentralblatt fur Bakteriologie, Mikrobiologie, und Hygiene. Cross experiments confirmed the high protective effect of the Pseudomonas common antigen (original endotoxin protein) contained in Pseudomonas vaccine, which, together with other soluble components, substantial afforded protection of mice against vibrios and thus confirmed the antigenic relationship between Pseudomonas and Vibrio. Satisfactory results were also obtained by vaccination with selected bacterial endotoxin lipopolysaccharides derived from some members of the Enterobacteriaceae family, which in some cases afforded 70-90% and in others only a 20-50% protection against Pseudomonas and Vibrio infection. The protective effect was reduced by up to 2/3 was obtained with vaccines subjected to alkaline hydrolysis. A single dose of Propionibacterium parvum cells not followed by specific vaccination afforded up to 50% protection against challenge with P. aeruginosa or V. parahaemolyticus. This decrease is evident regardless of whether the material is grouped according to serovars or to regions. However, the degree of resistance in multiresistant isolates increased. The frequency of S. saint-paul decreased, but continued to have the highest degree of resistance. In the D1 group there were almost exclusively mono-resistant isolates. 59% of all resistant isolates were resistant to tetracyclin, 47% to streptomycine and 25% to sulfonamides. Sensitive isolates predominate in outbreaks, except for one outbreak of S. enteritidis, which showed a marked percentage of bacteria with reduced sensibility against cephalosporine. The resistance patterns showed considerable variance over the time of observation. As bacteria may be involved in the carcinogenic process, we investigated the effect of cholecystectomy and dimethylhydrazine (DMH) administration to SPF NMRI mice with regard to tumour genesis and bacterial colonisation of the intestine. It results from this study that cholecystectomy does not influence tumour genesis and that 6-7 months post operationem and DMH administration tumours and bacteria originally not found in the animals develop: clostridia, eubacteria spec. which cannot be differentiated and E. lentum. Theses changes appear in group II of mice (laparotomy and DMH) and group III (cholecystectomy and DMH), but not in group I (controls). From the results of this study we cannot conclude whether the tumours or the new bacteria appeared first. Biochemical investigations of C. innocuum, C. paraputrificum and C. tertium indicated that these bacteria metabolised bile acids by a specific metabolic step only but not produced carcinogenic substances themselves. If bacteria are involved in tumorgenesis, different species may be involved producing a carcinogenic environment by metabolic chain reactions. The reliability of the Api 20A system in identifying Clostridium species was studied by performing biochemical tests both in the microsystem and according to the VPI Anaerobic Handbook. 66% of the Clostridium strains could be identified by the Api 20A system supplemented, in accordance with the Api Handbook by other tests such as morphology, lipase and lecithinase production. The following Clostridium species, known as clinically most significant, could be identified by the Api 20A system: Clostridium sordellii and C. sporogenes (92%), C. bifermentans (60%) and C. tertium (78%). Methods which can be used in the clinical laboratory for the identification of Clostridium species are described, i.e. isolation of Clostridium strains from mixed cultures, examination of morphological characteristics, performance of biochemical tests in the Api 20 A system as a micromethod, verification by conventional biochemical test systems and gaschromatographic analysis in special laboratories if necessary.</p>","PeriodicalId":23821,"journal":{"name":"Zentralblatt fur Bakteriologie, Mikrobiologie, und Hygiene. {"title":"[Distribution of species of the genus Listeria].","authors":"J Rocourt,&nbsp;H P Seeliger","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Within the classification of different Listeria species according to present knowledge, this study investigates the sources and distribution of both the known \"classical\" species of Listeria as well as the recently added species: L. monocytogenes, L. innocua, L. ivanovii, L. welshimeri, L. seeligeri, L. grayi and L. murrayi. For this purpose 573 isolates from the environment, food, healthy carriers and patients suffering from listeriosis were selected out of a total of approximately 6000 strains originating from all parts of the globe which are maintained at the Listeria Culture Collection of the Institute of Hygiene, Würzburg. The selection was made according to the criterium of widest possible geographic distribution. The results indicate the environment to be the natural reservoir of all of the 7 'new' Listeria species. Healthy animal carriers harbour in their intestinal tract all species of Listeria with the exception of L. murrayi, whilst in the excreta of healthy individuals only the species L. monocytogenes, L. ivanovii, and L. innocua were found. Identification of strains from pathological specimens of human and animal origin resulted invariably in the finding of L. monocytogenes or L. ivanovii. L. grayi (7 strains) was found sporadically in Europe and in the USA, whereas L. murrayi (9 strains) has been isolated only in the USA. 26 of 28 so far known strains of L. welshimeri originated from the USA, and only 2 strains of this species were found in Europe. L. seeligeri (80 strains) was identified among European isolates, L. innocua, however, in both USA and Europe. L. ivanovii occurs in Australia, Europe and the USA. L. monocytogenes is of global distribution. Table 1 presents a survey of species, serovariety and origin of the 573 isolates. {"title":"[Immunity of the skin--infection and percutaneous immunization of rabbits with E. coli ATCC 13676].","authors":"W Hammerschmidt,&nbsp;E Hellmann","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>After intracutaneous infection of rabbits with a suspension of E. coli which was followed by a transient local inflammation, the local and systemic immune responses were determined using the lymphocyte stimulation test (LTT) and the hemolysis plaque assay (HPA). Lymphocytes of the lymphatic system draining the infected skin area and blood lymphocytes were used. With lymphocytes derived from the local lymph nodes, a substantial increase of specific stimulation in the LTT was detected beginning at day 3 after infection and lasting up to the termination of the experiment (3 weeks). Blood lymphocytes were stimulated at a lower level: The activity showed a peak at day 4 and an elevated level only during a 10-day period. After the intracutaneous infection with E. coli, increasing numbers of antibody-releasing lymph node cells were detected in the HPA. The antibody-secreting cells of the IgM and IgG classes clearly showed an increasing specificity for E. coli lipopolysaccharide coupled to sheep red blood cells. As with the LTT, the highest activities (values of specificity and number of plaque-forming lymphocytes) were observed at the end of the experimental period. An emulsified preparation of a heat-inactivated E. coli culture (E. coli-BKS) which had been applied locally onto the artificially altered skin evoked a similar immunological response after a 2 or 3-weeks treatment. In such animals an increased activity of lymph node cells could be registered by LTT and HPA as compared to reactions from placebo-treated control animals. However, the topical immunization with nonviable E. coli stimulated not only lymphocytes which produced antibodies directed specifically against E. coli lipopolysaccharide as demonstrated by the HPA. An increased number of lymphocytes reacted even with native sheep red blood cells. The microscopical examination of samples after staining with acridine-orange has been found to be a rapid and simple method, which may reduce in 88% of the samples the time required for the processing and thus increase the value of blood-cultures for the treatment of patients with bacteremia significantly. The determinations were carried out in a culture test on agarose gel and in a liquid test system (turbidity test according to DiFerrante). The results of both methods used showed that the three Streptococcus species could be differentiated by the relative quantitative determination of hyaluronidase activity. S. equisimilis strains produce 5 to 10 times more hyaluronidase than those of S. zooepidemicus. The strains of S. equi did not show enzyme production. In addition parallel tests by determination of final N-acetyl-D-glucosamine groups and by turbidity test were done to confirm the hyaluronidase activity of S. equisimilis strains.
