Jawahar","doi":"10.2306/scienceasia1513-1874.2022.005","DOIUrl":"https://doi.org/10.2306/scienceasia1513-1874.2022.005","url":null,"abstract":"The growth and composition of periphyton in low saline groundwater (5 ppt) were evaluated using 5 different substrates: split bamboo poles (SBP), green shade net (GSN), high density polyethylene (HDPE) sheets, low density polyethylene (LDPE) sheets, and polyvinyl chloride (PVC) pipes). The effect of substrate type, submersion time, and depth was evaluated. The average submersion depth of all the substrates was maintained at 88.27 cm, and this trial was conducted for 60 days. Chlorophyll a, dry weight, ash, ash free dry weight, and periphyton diversity were analyzed. The growth of periphyton in terms of chlorophyll a value on different substrates has revealed a higher mean value for bamboo substrate (6.82±2.79 μg/cm2), followed by green shade net, HDPE, LDPE, and PVC with values of 5.24±2.27 μg/cm2, 4.64±2.06 μg/cm2, 4.51±2.08 μg/cm2, and 3.10±1.57 μg/cm2, respectively. The mean values of dry weight recorded were 1.81±0.78 mg/cm2, 1.65±0.70 mg/cm2, 1.29±0.56 mg/cm2, 1.32±0.57 mg/cm2, and 0.78±0.45 mg/cm2 for SBP, GSN, LDPE, HDPE, and PVC substrates, respectively. In all substrates, there was a significant difference observed in the development of periphyton over time (p < 0.05). MicroRNA (miR)-21a-5p and Smad7 have been reported to play an important role in the fibrosis process. However, how miR-21a-5p involves in the TGF-β1-induced myofibroblasts transformation of keratocytes remains unclear. In the present study, we found an increase in the expressions of α-SMA and collagen I at protein level as well as Smad7 and miR-21a-5p at mRNA level when keratocytes were treated with TGF-β1Here; but the Smad7 at protein level kept unchanged. The overexpression of miR-21a-5p attenuated Smad7 protein expression, and miR-21a-5p inhibition promoted Smad7 protein expression without affecting its mRNA expression. Furthermore, inhibition of miR-21a-5p could decrease TGF-β1-inducedα-SMA expression, whereas the addition of Smad7 knockdown would rescue the expression of α-SMA. In our previous work, treatment with the combined antimicrobials has been shown to significantly inhibit this polyspecies biofilm; however, the proteomic changes associated with this biofilm inhibition remain not well understood. Thus, the present work was performed to determine the proteome profile of P. aeruginosa-C. albicans polyspecies biofilms following treatment with the combined antimicrobials. The P. aeruginosa-C. albicans polyspecies biofilms were developed in 6-well microplates at 37 °C for 24 h under aerobic and anaerobic conditions. Following the treatment with 40 mg/ml combined antimicrobials (20 mg/ml erythromycin:20 mg/ml nystatin), whole-cell proteomes from the polyspecies biofilms were analyzed by a combination of two-dimensional polyacrylamide gel electrophoresis (2D SDSPAGE) and mass spectrometry. On the other hand, the differentially expressed proteins were analyzed using STRING database. Results demonstrated that 3 and 9 P. aeruginosa proteins were differentially expressed in treated polyspecies biofilms under aerobic and anaerobic conditions, respectively. Based on the protein interaction network, several biological pathways such as carbohydrate metabolism, protein metabolism, and secondary metabolite metabolism in P. aeruginosa might be affected by the treatment with combined antimicrobials. There were no differentially expressed C. albicans proteins identified herein. It is possible that the inhibition of the polyspecies biofilms by the combined antimicrobial is associated with multiple biological pathways. The molecular mechanism of OSCC progression remains unclear. The aim of this study was to explore specific prognostic biomarkers of OSCC at the transcriptome-wide level. A total of 158 differentially expressed long non-coding RNAs (lncRNAs), including 85 cis-acting and 10 trans-acting differentially expressed lncRNAs, were screened in OSCC tissues compared with adjacent normal tissues. Both cisand trans-target genes of differentially expressed lncRNAs were enriched in pathways associated with cancer progression. Additionally, 25 differentially expressed mRNAs were screened. Overlapping analysis of differential protein-coding genes with the cisand trans-target genes of differentially expressed lncRNAs indicated that KIF19 was the only trans-acting gene of differentially expressed lncRNA ‘ENST00000626052’ (named ENSTa). ENSTa was down-regulated in OSCC tissues and cells, and its low expression corresponded to a poor prognosis in OSCC. KIF19 was highly expressed in OSCC tissues and cells. Patients with high expression of KIF19 had a poor prognosis. Cellular experiments showed that KIF19 expression and cell proliferation were inhibited in cells overexpressing ENSTa. However, KIF19 overexpression counteracted the effect of ENSTa overexpression on cell proliferation. The physical and chemical characteristics of MA-AC were evaluated by N 2 adsorption / desorption, Scanning Electron Microscope (SEM), Fourier Transform Infrared (FTIR), and X-ray Photoelectron Spectroscopy (XPS). The results indicated that the MA-AC has more microporous structures and more oxygen bearing functional groups than the unmodified AC (denoted as AC). The Ni 2 + adsorption behaviors of the adsorbents were investigated by batch experiments, which indicated that the MA-AC showed 38% higher Ni 2 + adsorption capacity than the unmodified AC. The adsorption isotherms and the kinetics of the absorbents agreed well with the Langmuir model and the pseudo-second-order model. The Ni 2 + adsorption mechanisms of MA-AC were investigated by FTIR and XPS analyses.
