Abstract

The concerns raised by the authors of this commentary are largely addressed in the paper and the requested information is available in the supplementary materials and the published trial protocol referenced in the paper. We have further detailed some information to address their concerns. The planned effect size was based on the results reported by the largest randomized controlled trial at the start of our trial and supported by peer review and patient and clinician input. The trial was powered for the primary outcome of live birth, which we have reported. All reported secondary outcomes and subgroup analyses are only hypothesis generating.

Context: The period of human embryonic growth covered is between the formation of the primordial germ cells in the fetus until mid-pregnancy. High quality research on lower animals is included if it helps to clarify the human situation. Studies progressing to birth and later are published if they have a direct bearing on events in the earlier stages of pregnancy.
