Abstract
The absolute configuration of cubensic acid was predicted using the “Biochemistry-based Rule” and confirmed through total synthesis. Key steps in the synthesis included a titanium-mediated aldol reaction, a Paterson anti-aldol reaction, and a vinylogous Mukaiyama aldol reaction.
