Abstract

Vicinal dithiol-containing proteins (VDPs) are a special type of redox-active thiol-carrying ubiquitous proteins with essential role in maintaining redox balance. The abnormal cellular levels of VDPs are known to be implicated in numerous pathological conditions including cancer, diabetes, stroke and neurodegenerative diseases. Therefore, developing small molecule fluorescent tools for the sensitive and selective detection of VDPs is indispensable. Herein, we rationally designed and screened a red-emitting fluorescent probe (FRP-6) carrying α, β-unsaturated acrylamide as a receptor unit, for the precise detection of VDPs. The probe demonstrated excellent capability to identify its target both in the living cells and in vivo without being interfered by other biologically relevant analytes. Additionally, FRP-6 was employed to detect the depletion of VDPs levels in drug induced liver injury (DILI) and acute alcoholic liver injury (AALI) models.
